An enzyme immunoassay (EIA) for the quantitative determination of insulin-like growth factor binding protein-3 (BP3) was established, and reported herein. BP3 was purified from pooled human serum by column chromatography with Superose^<TM>10/30,HiTrap Heparin and antibody bound Sepharose 4B. The molecular weights of the purified BP3 were 150 kD, 53 kD and 47 kD. The antigenicity of the purified BP3 was equal to that of the commercially obtained recombinant BP3. The growth rat cultured mesangial cells 10^4 mesangial cells per milliliter was accelerated by human recombinant IGF-I, with maximum acceleration obtained at 12.5ng/ml and inhibeted by human recombinant BP3 with maximum inhibition obtained 1.0mg/ml. The serum levels of BP3 in normal children were correlated to their serum levels of creatinine (R=0.61,p=0.004). However, there was a weak correlation between the serum levels of BP3 and those of creatinine in children with renal disease, whose serum creatinine levels exceeded 1.0mg/dl (R=0.493,p=0.123). The serum IGF-I levels were also correlated to those of creatinine in normal children, while there was no correlation between the levels of serum IGF-I and serum creatinine in children with renal diseases. The serum level of unsaturated IGF-I binding protein (UBP) decreased according with the decrease in the amount of excreted urinary protein (R=-0.361,p=0.118).
