The effects of interleukin 6 (IL-6) on gene expression of both β_1 and α_5 subunits of integrin in rabbit corneal epithelial cells were investigated by the reverse transcription-poly-merase chain reaction (RT-PCR). Since the cDNA sequence of integrin β_1 and integrin α_5 of the rabbit have not been reported, the cDNAs of the β_1 and α_5 subunits of integrin were prepared from cultured corneal epithelial cells of the rabbit and the DNA sequence of rabbit β_1 and α_5 integrin was determined. Rabbit cDNA for integrin β_1 showed 95.2% homology to both human and mouse integrin β_1. The homology of integrin α_5 was 93.0% to human integrin α_5 and 88.9% to mouse integrin α_5. When rabbit corneal epithelial cells were cultured with various concentrations of IL-6 (0.1-10.0ng/ml), the gene expression of integrin β_1 and integrin α_5 increased in a dose-dependent manner. The stimulatory effects of IL-6 on mRNA level for both β_1 and α_5 integrin reached the maximum at 1 hour after the addition of IL-6. Thereafter, the mRNA level decreased gradually. These findings indicate that IL-6 stimulates the gene expression of integrin α_5β_1 (fibronectin receptor) in rabbit corneal epithelial cells.
