The effects of histaminergic agonists on the production of interleukin-6 (IL-6) and cell proliferation in endothelial cells and synovial fibroblastic cells were examined. Both histamine and H_1-receptor agonist, 2-pyridylethylamine significantly increased IL-6 production and cell proliferation in both types of cell, while H_2-receptor agonist, dimaprit significantly decreased IL-6 production and cell proliferation in both types of cell. Serum and synovial fluid were obtained from patients with rheumatoid arthritis (RA) or osteoarthritis (DA), and their effects on the production of IL-6 and cell proliferation in endothelial cells and synovial fibroblastic cells were examined. Synovial fluid from RA patients significantly increased these events in both types of cell, without stimulation by histaminergic agonists. In addition, synovial fluid from RA patients suppressed the effects of the H_2-receptor agonist on IL-6 production and cell proliferation both in endothelial cells and synovial fibroblastic cells. These findings indicate that a decrease in the suppressive effects of H_2-histaminergic agonist on IL-6 production and cell proliferation both in endothelial cells and synovial fibroblastic cells's involved in perpetuating the inflammatory process in RA.
