The W^<sh> is a mutant allele at the W (c-kit) locus of mice. Mice of W^<sh>/W^<sh> genotype have white hair and black eyes. Although adult C57BL/6-W^<sh>/W^<sh> mice are not anemic, they showed a marked depletion of mast cells. Most homozygous or double heterozygous mice at the W (c-kit) locus whose mast cell depletion was comparable to that of W^<sh>/W^<sh> mice are sterile due to depletion of germ cells, but male and female W^<sh>/W^<sh> mice have many germ cells in their gonads. We investigated the mechanism of mast cell depletion of W^<sh>/W^<sh> mice. Cultured mast cells (CMC) derived from the spleen of W^<sh>/W^<sh> mice did not attach to normal (+/+) fibroblasts and disappeared within 2 weeks after the initiation of the co-culture. W^<sh>/W^<sh> CMC did not express the extracellular domain of c-kit protein and moreover did not contain c-kit protein. The c-kit mRNA was strongly expressed in +/+ CMC but was not detectable in W^<sh>/W^<sh> CMC. In spite of the lack of c-kit mRNA in W^<sh>/W^<sh> CMC, c-kit mRNA was detectable in the spleen, testis and cerebellum of W^<sh>/W^<sh> mice. No significant changes were found in the coding region of c-kit transcripts obtained from the cerebellum of W^<sh>/W^<sh> mice. Taken together, the expression of the c-kit gene in W^<sh>/W^<sh> mice appeared to be influenced by the type of cell ; it was normal in the cerebellum, weak in the testis and spleen, and not detectable in CMC. The symptoms of C57BL/6-W^<sh>/W^<sh> mice paralleled the expression of c-kit gene.
