<原著>ヒスタミン受容体作用薬のマウスリンパ球抗体産生及びコラーゲン関節炎に対する影響

This study was carried out to investigate the role of histamine receptors in IgG production by spleen cells and the effects of histaminergic agonists on type II collagen-induced arthritis in mice. Con A-derived IgG production in the spleen cells from normal DBA/1J mice was dose-dependently suppressed by histamine. Moreover, both of histaminergic H_1 and H_2 agonists suppressed Con A-derived IgG production in the normal spleen cells. In CIA mice, spontaneous IgG production in the spleen cells was significantly higher than that in normal mice, but almost no Con A-derived production was observed in the spleen cells. Histamine induced suppression of the spontaneous IgG production in the spleen ceils with CIA was weaker than the histamine-induced suppression on Con A-derived IgG production in the normal spleen cells. The H_2-agonist-induced suppression on the IgG production in the CIA spleen cells was weaker than that in the normal spleen cells. On the other hand. the H_1-agonist-induced suppression on the IgG production in the CIA spleen cells was stronger than that in the normal spleen cells. Both H_1 and H_2 histaminergic agonists suppressed the incidence of onset with arthritis, arthritic index. and the titer of the anti-type II collagen antibody in CIA mice. The H_1 agonist had stronger benefical effects than the H_2 agorist. These findings suggested that both H_1 and H_2 histaminergic agonists had suppressive effects on in vitro IgG production in the spleen cells and also on in vivo disease activity in CIA.
