Urokinase type plasminogen activator (u-PA) digests fibronectin and is postulated to play an important role in cell migration. In the present paper, the role of u-PA in corneal epithelial migration was investigated using organ cultured rabbit corneas and dish cultured epithelial cells. Significant correlation was observed between the length of the path of the corneal epithelial migration and plasminogen activator (PA) activity in the culture medium. Zymography revealed that the PA in the culture medium was of the urokinase type. Immunohisto-chemical study showed that u-PA and plasminogen were present in the leading edge of the migrating epithelium in the cultured corneal blocks. PA activity of epithelial cells was extremely high in the culture medium as compared with that in keratocytes or endothelial cells. The addition of anti-human u-PA retarded the migration of corneal epithelium in a dose dependent fashion. Wounding of the confluent corneal epithelial cell cultures increased PA activity in the culture medium. These results suggest the active involvemint of u-PA in the process of corneal epithelial resurfacing and tissue remodeling. It was suggested that alternating occurrence of cell attachment to fibronectin and cell detachment by degradation of fibronectin by u-PA play an important role in the motility or migration of corneal epithelial cells.
