Plasminogen activator (PA) in the cellular components of bronchoalveolar lavage fluid (BALF) obtained from 50 patients was determined in order to clarify the source of PA in the airways. Alveolar macrophages (AM) in BALF were found to contain urokinase-type PA (u-PA), tissue-type PA (t-PA), or both types of PAs. When AM were cultured, AM secreted only u-PA into the culture medium, and the cell extract contained u-PA only. The influences of various pharmacological agents on PA secretion by cultured AM were investigated ; sulpyrine and hydrocortisone reduced PA secretion in culture medium, but OK-432,histamine, interferon-γ (IFN-γ) and platelet activating factor (PAF) increased PA activity in culture medium. PA activities in culture medium and cell extracts were decreased by the addition of actinomycin D, cycloheximide, and 5-fluorodeoxyuridine. In order to clarify the mechanism of secretion of u-PA by cultured AM, a model cell, U-937,a human monocyte-macrophage, was used. The secretion and synthesis of u-PA by U-937 were accelerated by PAF, IFN-γ or OK-432. U-937 possesses a receptor of PAF and IFN-γ, which may mediate the secretion of u-PA.
