Uptake of [^3H] choline and release of [^3H] acetylcholine (ACh) were examined in the synovial membrane of the rat knee joint. Kinetic analysis of [^3H] choline uptake revealed two uptake systems, a high affinity component with a Km of 3.1×10^<-6>M and a low affinity one with a Km of 66×10^<-6>M. [^3H] Choline uptake was inhibited by removal of external Na^+ or treatment with hemicholinium-3. High K^+ evoked release of [^3H] ACh from the membrane preloaded with [^3H] choline was Ca^<2+>-dependent and Mg^<2+>-sensitive. γ-Aminobutyric acid (GABA) and GABA_A receptor agonist (muscimol) evoked [^3H] ACh releases were inhibited by bicuculline. High K^+ evoked [^3H] ACh release was inhibited by GABA_B receptor agonist (baclofen). These results suggest the existences of cholinergic neurons and GABA receptor in the rat synovial membranes, and interaction of the two.
