An IgG_1 murine monoclonal antibody (KA-1) to human keratinocyte cell membrane was prepared from BALB/c mice immunized with cell suspensions of both human epidermoid carcinoma cells (A431) and epidermal cells from psoriatic plaques. In frozen sections of normal human skin immunofluorescent staining techniques showed KA-1 stains in the epidermal cell membrane from the basal layer to the stratum granulosum, but not in the cell membrane toward the basement membrane and the cell membrane in the stratum corneum. On immunoblot analysis, KA-1 reacted with a 44 kilodalton protein extracted from both A431 cells and cultured human keratinocytes with Nonidet P-40. Immunofluorescent staining of epidermal monolayer cells grown on the coverslips showed punctate staining along the cell-to-cell contact surfaces, but not on the free surfaces. When dispersed A431 cells and epidermal cells were preincubated with various concentrations of KA-1 for 1 h at 4℃ before plating, the inhibition of cell attachment on the surfaces of 96-well flat-bottom microculture plates occurred with 0.1 μg protein/ml or more of KA-1. This inhibition was concentration-dependent, and not due to an immediate toxic effect of KA-1. Other murine monoclonal antibodies : anti-epidermal growth factor receptor, anti-cytokeratins, anti-human T cell (CD3), anti-actin and anti-vinculin, did not inhibit the attachment of cells. These results suggest that KA-1 recognizes the keratinocyte membrane antigen associated with cell attachment.
