An uricase was extracted by freezing, thawing and autolysis from Candida utilis grown on a medium contained of uric acid . The uricase was purified by ammonium sulfate fractionation (0.3-0.5 saturation), Sephadex G-100 and DEAE-cellulose column chromatography. The enzyme was purified about 80-fold by this procedure from original extracts. The enzymatic properties of the purified enzyme was studied . Optimum pH was 8.0,stable pH range was from 7.0 to 11.0 and optimum temperature was 35℃. The activity was inhibited by Cu^<2+>, Mn^<2+>, CN-and PCMB, while the activity was activated by glutathion. The lyophilized enzyme was stable at room temperature for a long time.
