イネcaffeic acid 3-O-methyltransferaseのクローニングおよび機能解析

イネファイトアレキシンの一つであるサクラネチンの生合成酵素naringenin 7-O-methyltransferase (NOMT)の既に報告されている部分アミノ酸配列を用いて, UV処理したイネ葉のcDNAからO-methyltransferase cDNAをクローニングした.しかし, 大腸菌で発現した融合たんぱく質(約41kDa)はNOMT活性を示さず, caffeic acid 3-O-methyltransferaseの活性を示した.定量RT-PCRの結果, その遺伝子は発芽後1-5週間のイネで構成的に発現しており, 塩化銅, ジャスモン酸及びUV処理による誘導性はないことが明らかとなった.基質特異性を調べた結果, B環に隣接している水酸基を二つ持つフラボノイド系化合物が良い基質となることが分かった.これらの結果から, 以前に報告されたNOMTの部分アミノ酸配列はNOMTではなく, caffeic acid 3-O-methyltransferaseのものであることが明らかとなった.
