We performed X-ray diffraction, cytological and histopathological studies to investigate biological affinities when different temperatures were used for sintering α-TCP, the major component of hydraulic calcium phosphate cement (CaPcement) that solidifies utilizing hydrolysis of α-TCP, DCPD and TeCP. The temperature for sintering α-TCP was set to 1, 200, 1, 300 and 1, 400℃. Transforming α-TCP into hydroxyapatite was slower in sintering temperature of α-TCP. In cytological study, growth rates of osteoblast-like cells on cement were investigated. Mouse-derived osteoblast-like cells, MC3T3-E1, was inoculated on about 0.2 mg CaPcement at 1×10^5 cells per well and culutured in α-MEM, then growth rates on days 3, 7, 14 and 21 were measured. Further, morphologies of the cells in growth phase and surfaces of the cement under the same condition were observed under a scanning electron microscope. On histopathological study, cavities with a 3 mm diameter were formed in the tibia and mandible in 30 rabbits weighing 3 kg and sclerotic CaPcements with different sintering temperatures were packed into these cavities. After 1, 3, 6 and 12 months, paraffin-embedded sections were prepared and stained with HE, then observed under a light microscope. Results were as follows. I. The difference of transforming α-TCP into hydroxyapatite. Transforming α-TCP into hydroxyapatite was slower in sintering temperature of α-TCP. II. Cytological observation A. Cell growth seemed slightly better in the group cultured on cement with a sintering temperature of 1, 300℃, but the differences between the three groups disappeared after incubation for 21 days. B. There was no difference observed in cell morphology between three groups. Satisfactory colony formations were observed after incubation in all three groups. III. Histopathological observations A. In the tibia, osteogenesis was observed at the periosteum, marginal and bone marrow sides of the cements after one month in three groups. However, after three months, the new bones matured and the margins were irregular. The new bones at the bone marrow side surrounded Hardening Materials. Marked differences due to differences in the sintering temperature were not found during these observations. B. In the mandibula, features similar to those of the tibia were observed and there were no marked differences due to differences in the sintering temperature. The above results indicated that temperature of hydraulic calcium phosphate cement does not markedly influence osteoblasts or osteogenesis.
