The effect of normal and high calcium supplementation on endochondral ossification at metaphysis of tibia was investigated, using male Wistar rats aged 5 weeks corresponding to early childhood in human beings, to which low calcium dietary had been given to result in bone loss. The results were reported as follows. I. Histopathologic findings In contrast to the control groups, the low calcium diet groups showed expanded chondrocyte layer, sparsely arranged hypertrophic layer, and decreased calcified matrix and its erosion. Compared with the low calcium diet normal diet groups, the low calcium diet mixed diet groups showed decreased width of chondrocytic layer, increased calcified matrix and its erosion, implicating promoted endochondral ossification. II. Scanning electron microscope findings The low calcium diet groups showed indistinct chondrocytic lacunae, less fusion of calcified vesicles, and expanded bone resorption in subchondral layer of bone formation, compared with the control groups. Compared with the low calcium diet normal diet groups, the low calcium diet mixed diet groups showed clearcut chondrocytic lacunae, more fused calcified vesicles, less bone resorption, suggestive of bone construction facilitated. III. Enzyme histochemical findings The low calcium diet groups showed less alkaline phosphatase (ALP) positive cells on trabeculae continuous with the new-formed trabeculae. ALP positive cells in the low calcium diet normal diet groups and the low calcium diet mixed diet groups were revealed identical to those in the control groups. Tartrate resistant acid phosphatase (TRAP) positive cells were the most in the low calcium diet groups, less in the low calcium diet normal diet groups, the least in the low calcium diet mixed diet groups, which were comparable to the control groups. As indicated above, functional balance and formation of bone matrix were facilitated with sufficient intake of calcium, in circumstances of bone loss, in adolescent years.
