In the junior high school science, soil microorganisms, such as fungi and bacteria, are dealt with as the reducer in the ecosystem. However, in the experiment utilizing these microorganisms, the starch is always used as an organic matter. In order to understand these microorganisms appropriately as the reducer, it is necessary to use the cellulose as an organic matter. The purposes of this study are twofold: to clarify the conditions, such as kinds of the optimal soil layer, paper, and culture days for cellulose-decomposition by soil microorganisms. The results were as follows: 1) as soil condition, the cellulose-decomposition turned out to be the strongest at the Ao layer in both the broadleaf forest and the coniferous forest, 2) as paper condition, the sugar concentration generated from recycled paper and newspaper was the almost same, 3) the highest sugar concentration was the 4th day from the onset of the culture.
