Effect of hydrostatic pressure on the reactions catalyzed by β-Galactosidase in eitherfree soluble state or immobilized on polymer micro beads was investigated. Both thehydrolysis of lactose to glucose and galactose and the formation of oligomer such asGal-Gal-Glc were investigated. Up to 150-200 MPa this enzyme showed only a smallirreversible inactivation,as detected by the activity assay after incubation at elevatedpressure and by the in situ fluoresce ncemesurement under the elevated pressure. Ineither free and immobilized state,the hydrolytic reaction was accelerated. This indicatesthe negative activation volume of the nucleophilic attack of water on galactosyl-enzyme(covalent) complex. The oligomer formation by free enzyme was depressed by increasingpressure but slightly higher yield of oligomer was observed for the immobilized enzyme under high pressure.
