The effect of intragastric gavage administration of docosahexaenoic acid (DHA) on 1, 2-dimethylhydrazine (DMH) -induced aberrant crypt foci formation in the rat colon was investigated. Male F344 rats were treated three times s. c. with 20 mg/kg of DMH and were given intragastrically either 0.7 ml of DHA or water 5 times a week for 4 weeks from the day before the first carcinogen treatment. The total number of DMH-induced aberrant crypt foci per colon was significantly less in the group given DHA than in the control group (42.4±18.7 vs. 122.1±35.3 ; p<0.001). The mean size of aberrant crypt foci, measured as the number of crypts per focus, was significantly smaller in the group given DHA than that in the control group (1.60±0.20 vs. 1.88±0.22 ; p<0.01). The results suggest that DHA suppresses the formation and the growth of aberrant crypt foci and may have a preventive effect on colon carcinogenesis.
