Epidermal growth factor receptor (EGFR) is a glycoprotein with a molecular weight 170 kilodaltons, and is reported that overexpression of EGFR was found more often in squamous cell carcinoma than in adenocarcinoma. In this study, the expression of EGFR was exmined by ^<125>I-EGF binding assay in 50 cases of gastric cancer, 7 gastric acancer xenografts in nude mice, 8 cases of esophageal cancer and 18 cases of colonic cancer. Western blotting and immunohistochemistry for EGFR were also done by using anti EGFR antiboby in gastric cancer xenografts. EGFR was detected in 50.0% (25/50) of gastric cancer, 87.5% (7/8) of esophageal cancer and 61.1% (11/18) of colonic cancer. Average EGF binding capactiy of EGFR-positive cases was 382±30.8 fmol/mg・protein (mean±SE) in gastric cancer, 770±34.7 in esophageal cancer and 61.9±5.9 in colonic cancer. EGFR was also prsent in 24.0% (12/50) of non-cancerous gastric mucosa, 100% (8/8) of esophageal mucosa and 44.4% (8/18) of colonic mucosa, however EGF binding capacities of non-cancerous mucosa of three tissues were lower than those in cancerous tissues. Average EGF-binding capactiy was 55.6±6.0 in gastric mucosa, 132±7.9 in esophageal mucosa and 64, 4±5.1 in colonic mucosa. All of the gastric cancer xenografts in nude mice showed EGFR-positive not only in binding assay but also in Western blotting and immunohistochemistry. There was a negative correlation between EGF binding capacity and tumor doubling time (p<0.01).
