Optimum Media for Outdoor Large-scale and Indoor Small-scale Batch Style Culture of <I>Nannochloropsis oculata</I>

A new enriched seawater medium for <I>Nannochloropsis</I> (ESM-NA medium) using analytical grade inorganic salts for indoor small-scale culture of <I>Nannochloropsis oculata</I> was formulated by removing ineffective chemicals as nutrients from Guillard F medium. In addition, a fertilized seawater medium for outdoor large-scale culture of the alga (FSM-NA medium) was devised in consideration of cost reduction. The composition of ESM-NA medium was NaNO<SUB>3</SUB>; 4.41 mM, NaH<SUB>2</SUB>PO<SUB>4</SUB> • H<SUB>2</SUB>O; 90μM, Fe-EDTA; 29.7μM, MnCl<SUB>2</SUB> • 4H<SUB>2</SUB>O; 2.2μM, ZnSO<SUB>4</SUB> • 7H<SUB>2</SUB>O; 0.38μM and CoCl<SUB>2</SUB> • 6H<SUB>2</SUB>O; 0.126μM, while that of FSM-NA medium was KNO<SUB>3</SUB>; 5.3 mM, NH<SUB>4</SUB>H<SUB>2</SUB>PO<SUB>4</SUB>; 0.66 mM, Clewat 32; 20 mg/<I>l</I> and Clewat Fe; 3 mg/<I>l</I>. The growth rate of <I>N. oculata</I> in FSM-NA medium was higher than that in YASHIMA medium which has been commonly used in Japanese hatcheries. Moreover, it can be considered that the contamination of FSM-NA medium into larval rearing water at the same time as the supplementation of <I>N. oculata</I> was little effect to the survival and the growth of fish larvae. Therefore, FSM-NA and ESM-NA media seem to be optimal for outdoor large-scale and indoor small-scale culture of <I>N. oculata</I>, respectively.
