We conducted pharmacokinetic studies after oral administration of 250 mg metformin to reexamine its dose-setting in Japan, where the maximum dose is limited to 750 mg daily.Tmax was 4 hrs in healthy controls (n=6) and diabetic patients (n=5).Cmax was 0.54±0.15μg/m<I>l</I> in control and 0.85±0.19 μg/m<I>l</I> in diabetic patients, consistent with data reported in the English literature. In repetitive administration of 500 mg metformin daily for 7 days, serum concentration of metformin reached the steady state on day 3, and the profile of metformin concentration on day 7 fitted well with the concentration curve predicted by that on day 1.No significantdifferences were seen in Tmax, Cmax, or T1/2 on days 1 and 7.We report the pharmacokinetics of metformin at the dose set in Japan.
