Sequencing-based typing (SBT) facilitates DNA typing of HLA. Using the Match Maker and MT Navigator in combination is particularly useful for rapid <I>DQA 1, DQB 1</I>, and <I>DRB 1</I> typing, enabling larger numbers of specimens to be rapidly and accurately typed.<BR>To determine HLA-associated genetic factors in Japanese children with type 1 diabetes, we analyzed 166 patients with acute onset type 1 diabetes.Patients were divided into 3 groups by age at onset, i.e., 0-4, 5-9, and 10-15 years.Among them, significant positive association was seen in <I>DQA 1*03-DQB 1*0303-DRB 1*0901, DQA 1*03-DQB 1*0401-DRB 1*0405</I> and <I>DQA 1*03-DQB 1*0302-DRB I*0802</I> haplotypes and negative association in <I>DQA 1*03-DQB 1*0302-DRB 1*0403, DQA 1*0102-DQB 1*0602-DRB 1*1501, DQA 1*0103-DQB 1*0601-DRB 1*1502</I> and <I>DQA 1*0103-DQB 1*0601-DRB 1*0803</I> haplotypes. A significant positive association was seen in the <I>DQA 1*03-DQB 1*0303-DRB 1*0901</I> haplotype and at onset at 0-4 years.<BR>Analysis by age of onset suggests that, in addition to the difference in clinical course and the positive rate for islet autoantibodies, a difference may exist in the roles of HLA class II antigen genes in onset and/or progression of β-cell destruction.
