Study on Transforming Growth Factor .BETA. (TGF-.BETA.) mRNA in Renal Biopsy Specimens of Patients with Diabetic Nephropathy Using Non-radioactive in situ Hybridization.

This is the first report of non-radioactive in situ hybridization of transforming growth factor β(TGF-β) mRNA in renal biopsy specimens obtained from 13 patients with diabetic nephropathy (DM-N). In order to elucidate the localization of TGF-β mRNA in DM-N, we examined non-radioactive in situ hybridization in 13 renal tissues of patients with DM-N. The method used for this study has recently been developed in our institute using a 20 mer oligonucleotide probe. TGF-β mRNA positive cells were seen among mesangial cells and epithelial cells of the glomeruli, and the signals were strong in tissues with moderate mesangial matrix expansion but weak in nodular lesions. Moreover, we found that TGF-β mRNA positive cells were observed in atrophic tubuli and infiltrating cells of the interstitial area. It was concluded that TGF-β mRNA is expressed in renal tissues of patients with diabetic nephropathy, and that its expression may be related to the glomerular damage which characterizes this disease.
