Glucose metabolism in the conceptus during early organogenesis was investigated using cultured rat embryos at 10 and 11 gestational days.<BR>Rat concepti were incubated for 4 hours in 75% rat serum containing labeled (U-<SUP>14</SUP>C, 1-<SUP>14</SUP>C or 6-<SUP>14</SUP>C) glucose, and the metabolic fates of glucose were determined. Concepti on gestational day 10 were characterized by very high rates of glucose utillization and lactic acid production, and almost all utilized glucose was converted to lactic acid (89.63±2.78%). On day 11, the proportion of glycolysis significantly decreased (73.53±2.74%, p<0.01). The proportion of glucose oxidized was very low (0.75±0.04%) on day 10, but increased significantly (3.83±0.35%, p<0.01) on day 11. The relative contribution of the pentose cycle to glucose metabolism was 0.60±0.03% on day 10 and 1.41±0.05% on day 11.<BR>These results suggest that the conceptus on gestational day 10, before the esablishment of yolk sac circulation, is dependent on glycolysis and on gestational day 11, at the start of allantoic circulation, becomes less dependent on glycolysis, concomitant with the appearance of TCA cycle activity.
