To clarify the mechanisms of hypertriglyceridemia in NIDDM (non insulin dependent diabetes mellitus), we studied triglyceride secretion rates (TGSR) using Triton WR-1339 in NIDDM modelrats, prepared by the intraperitoneal injection of streptozotocin (STZ) at 2 or 5 days of age, andcompared them with those of adult diabetic rats given an intravenous injection of STZ (DM rats).We also studied the intestinal contribution to triglyceride metabolism by determining apoprotein Bisoproteins (% Bs in total apoprotein B) in triglycerdie-rich lipoproteins in the diabetic rats.<BR>Diabetic rats were divided into three groups according to fasting plarma glucose level (FPG) to study the relationship between hyperglycemia and triglyceride metabolism. Fasting triglyceride levels tended to increase in rats with FPG under 155 mg/d<I>l</I> and over 231 mg/d<I>l</I>, but not in other rats. TGSR of rats with FPG under 155 mg/d<I>l</I> was 0.54±0.03 mg/min, significantly higher than in control rats (0.45±0.02 mg/mm.). In rats with increased plasma glucose levels (more than 156 mg/dl), TGSR showed smaller values. The apoprotein Bs ratio (% Bs) as determined by the HPLC method was 14.4±1.2 % in triglyceride-rich lipoproteins from DM rats, significantly higher than that in control rats (8.8±1.5%). Insulin treatment improved %Bs in DM rats.<BR>These results indicate that hypertriglyceridemia in non-obese NIDDM model rats with mild elevation of FPG was accounted for in part by triglycerdie overproduction induced by hypoinsulinemia, and suggest that triglyceride-rich lipoproteins from intestine play a role in diabetic hypertriglyceridemia.
