Islet-cell surface antibodies and complement-dependent antibody-mediated cytotoxicity in the NOD mouse.

Islet-cell surface antibodies (ICSA) and complement-dependent antibody-mediated cytotoxicity (CAMC) were evaluated in sera from NOD mice, which have recently been described as an animal model of insulin-dependent diabetes mellitus (IDDM), and mouse anti-rat islet cell sera (MARICS).<BR>ICSA were measured by radioimmunoassay (RIA) using 125I-antimouse IgG antibody. A positive correlation was found between ICSA levels when measured using mouse islet cells or insulinoma cells (In-111) as target cells (r=0.79, p<0.01). Mean ICSA levels in sera from male and female NOD mice were both significantly higher than those in sera from control female ICR mice, but there were no significant differences in ICSA levels between male and female NOD mice. When ICSA levels in sera from male and female NOD mice with aging were compared, ICSA levels increased with age, reaching a plateau after 15 weeks of age.<BR>CAMC was measured using <SUP>15</SUP>Cr-labelled rat silet cells as target cells. Although CAMC levels in MARICS were significantly higher than those in control female BALB/C mice, control female ICR mice, male NOD mice and female NOD mice, there were no significant differences among control female BALB/C mice, control female ICR mice, male NOD mice and female NOD mice. When CAMC levels in sera from male and female NOD mice with aging were compared, CAMC levels at 20 and 27 weeks of age in female NOD mice were higher than those in male NOD mice. There were no correlations between ICSA levels and CAMC levels in sera from male and female NOD mice.<BR>The incidence of diabetes in female NOD mice was higher than male NOD mice. However, the incidence of the presence of ICSA was the same in male and female NOD mice in this study. These findings suggest that ICSA that are not related to cytotoxicity might exist. The elevation of CAMC levels at 20-27 weeks of age in female NOD mice may be related to the onset of overt diabetes.
