In the present study we investigated the cartinostatic primary effect and side effects of PP therapy by combined use of Cisplatin (CDDP) and Peplomycin (PEP) through intravenous and intra-arterial infusion.<BR>Intravenous infusion was performed in 11 patients (site of the primary lesion were the tongue, gingiva and buccal mucosa in 7, 2 and 2, respectively), and intra-arterial infusion in 13 (sites of the primary lesion were the tongue, gingiva, lip and floor of the mouth in 9, 2, 1 and 1, respectively). In the former route, CDDP 30mg/body/day and PEP 8mg/body/day were administered for 5 days each. In the latter route, CDDP 10 mg/body/day was continuous infused into the superficial temporal artery for 5 days, followed by administration of PEP 10mg/body/day for 5 consecutive days, recognized as one course (Kur). As a rule, two courses each of intravenous and intra-arterial infusion were performed.<BR>Response rate were 81.8%(including one CR, 8 PR, one MR and one NC) and 92.3%(including one CR, 11 PR and one MR) for the intravenous and intra-arterial infusion groups, respectively. The frequency of side effects of the intravenous infusion method was high, i. e., nausea in 100% and vomiting in 72.2%, whereas that for the intra-arterial infusion method was low, i. e., nausea in 46.2% and vomiting in 15.4%. There were no significant changes with time in erythrocyte or leukocyte counts or Ccr in either method. However, the platelet count significantly decreased as a result of intravenous infusion.
