The study of PGI2 producing activity by umbilical endothelial cells of toxemia of pregnancy.

It is suggested that low production of PGI2 by endothelial cells is one of aggravating factors in toxemia of pregnancy (TOX). Therefore, we investigated activity of PGI2 production by umbilical and maternal omental endothelial cells, and characterized their enzymatic activities.Stimulation of umbilical endothelial cells by thrombin (0, 0.1, 0.3, 0.5, 1.0iu/ml) brought about dose-dependent increases in PGI2 production in normal pregnancy (NOR) but not in TOX.Conversion of arachidonic acid to PGI2 (detected by 6-keto PGF1α) was shown to be significantly more efficient in umbilical and omental endothelial cells of NOR than in these of TOX.The apparent Km and Vmax values for the enzyme involved in PGI2 production in these cells were found to be significantly smaller in NOR than in TOX.Little difference in the apparent Km values of PGI2 synthetase was detected between NOR and TOX, whereas Vmax values were apparently increased in NOR.It is concluded that suppressed activity of cycloxygenase and probable phospholipase A2 may result in the decreased production of PGI2 in TOX.
