:A preliminary report on studies with murine leprosy developing in two mouse strains with diverse immunoreactivity

Thymus-dependent fibrin clearing has been demonstrated in granulomatous inflammation (Izaki et al., 1979). In this study we tried to detect plasminogen activator (PA) and its regulating inhibitor in two polar types of experimental murine leprosy. A susension containing 2×108 Mycobacterium lepraemurium was inoculated in the sternal area of C57BL and CBA mice. The lepromas were excised and homogenized in 0.05M Tris-HCl+0.1M NaCl, pH 7.5. PA activity and inhibitory effect for urokinase (UK) were assayed using PyroGlu-Gly-Arg-pNA and fibrin plates.C57BL mice developed a localized lesion at the site of inoculation. The lepromas obtained 4-6 weeks after infection exhibited inhibitory activity for UK. The lepromas obtained 8-11 weeks, on the other hand, demonstrated prominent activity of PA, whereas the UK inhibitor activity was no longer observed. The PA activity in tissue was shown associated with cutaneous ulceration with purulent discharge which was followed by the limitation of disease development.CBA mice developed a diffuse leproma, which progressively grew without the healing reaction. Such lepromas showed only a weak activity of UK inhibitor but no PA activity.Proteolytic activation and regulation of PA may relate to the development of murine lepromas and the defence mechanisms against the acid-fast bacilli.
