Acute, subacute and chronic toxicities of cefoxitin (CFX) were studied in experimental animals.<BR>The i. v. LD<SUB>50</SUB> value (g/kg) of CFX was 5.10 in mice, 8.98 in rats and>10.0 in beagles. Cefazolin and cephalothin, reference drugs, gave respectively, 4.17 and 5.31 for i. v. LD<SUB>50</SUB> value (g/kg) in mice.<BR>In 30-day intravenous subacute toxicity study, soft stools, increased water intake and enlargement of cecum were observed in all of treated groups.<BR>In 6-month intravenous chronic toxicity study, soft stools were observed in the highest dose group, and increased water intake was seen in all of the treated groups. Urinalysis revealed a significant increase of urine volume, Na<SUP>+</SUP>, K<SUP>+</SUP>, Cl<SUP>-</SUP> and Al-P and decrease of urine osmotic pressure in the higher dose group. Pathologically, enlargement of the cecum was observed in most of the treated rats, and mild centrilobular fatty degeneration of the hepatocytes was found in 2 males receiving 1, 000mg/kg.
