Chemosensitivity of uterine cancer cell lines determined by the tetrazolium salt(MTT) assay.

Three human uterine cancer cell lines (OMC-1, 0MC-2, OMC-4) were cultured and their chemosensitivity to 13 antitumor agents (ACD, ADM, BLM, CBDCA, CDDP, CPM, 5-FU, MMC, MTX, PEP, VBL, VCR, VP-16) was tested by the 3- (4, 5-dimethylthiazol-2-y1) -2, 5-diphenyl tetrazolium bromide (MTT) assay. We have adopted the incubation times of 4 hr with 50μl of MTT (2mg/ml) added to 100 Jul medium and 200μl of dimethyl sulfoxide (DMSO) as the solvent of formazan. Good linearity was seen between optical density and cell number. OMC-1, 2, 4 cells were sensitive to actinomycin D, carboplatin, cisplatin, and 4-hydroperoxycyclophosphamide. OMC-1 was also sensitive to bleomycin, mitomycin C, and vinblastine. MTT assay is a useful in vitro test of chemosensitivity.
