The effect of premenopausal bilateral oophorectomy and of pseudomenopausal therapy with Gn-RH agonist on bone mineral content and bone metabolism.

Bone loss in women is thought to be mainly associated with declining estrogen level. In this study, the effect of premenopausal bilateral oophorectomy (PBO) and of pseudomenopausal therapy with Gn-RH agonist on bone mineral content and bone metabolism were evaluated. Bone mass was measured using dual photon absorptiometry (DPA). Bone mineral density (BMD) was found to be decreased to less than 2SD below the mean in 25% of patients with PBO, for whom BMD showed gradual decrease in comparison with that in age-matched controls. Decrease in total bone mineral content after six months was about 15% in the patients with PBO and about 11% in patients undergoing Gn-RH agonist therapy. Markers of bone metabolism showed high turnover, with levels of markers of both absorption and formation being elevated. However, levels of calcitonin and BGP decreased remarkably over a short period.
