Expression of renin, angiotensinogen, kininogen and kallikrein genes in the human placenta in pregnancy complicated with intrauterine growth retardation.

The expression of renin, angiotensinogen, kininogen and kallikrein genes in the term human placentae, obtained from 5 normal pregnancies, 5 toxemic pregnancies and 5 pregnancies complicated with intrauterine growth retardation (IUGR), has been examined by RNA blot hybridization analysis using the specific human complementary DNA (cDNA) probes. Plasma renin activity (PRA) in the maternal blood was also measured in these three groups as an index of the activity of systemic (renal) renin-angiotensin system (R-A system).<BR>Renin mRNA was detected in the placenta of all three groups, but angiotensinogen mRNA, kininogen mRNA and kallikrein mRNA were not detected. The total content of renin mRNA in the placenta from IUGR pregnancies was significantly lower than those from normal or toxemic pregnancies. There was no significant difference in the total content of renin mRNA between the normal and toxemic groups.<BR>On the other hand, there was no significant difference in PRA between normal and IUGR pregnancies. The toxemic group showed significantly lower PRA than the normal or IUGR group.<BR>These results and our previous findings, which suggest that local placental R-A system plays a role in regulating utero-placental circulation, may indicate that the decreased activity in the placental R-A system in IUGR pregnancy causes decreased utero-placental blood flow, which leads to fetal growth retardation.
