To elucidate if insulin acts directly on ovary, the possible effect of insulin on cultured porcine granulosa cells was investigated.Insulin (25mIU/ml) enhanced both basal and hCG-stimulated progesterone production by monolayer culture of porcine granulosa cells. Insulin was also found to be indespensable for FSH-stimulated induction of LH f hCG receptors. Furthermore, insulin promoted hCG-stimulated morphological luteinization, i. e. inauguration of epitheloid form, of porcine granulosa cells. Thus, it is evident that insulin plays a pivotal role in functional differentiation of porcine granulosa cells.Further studies on the mechanism of this direct effect of insulin revealed that porcine granulosa cells possess specific receptors for insulin with two classes of binding sites. Insulin increased glucose incorporation by cultured porcine granulosa cells and enhanced glycogen synthase activity of granulosa cells.From these results it will be concluded that insulin directly acts on granulosa cells via its specific receptors, first activates glucose metabolism, and then induces luteinization of porcine granulosa cells.
