It may be true that ovarian function is regulated by the hypothalamo-pituitary system. Nevertheless, corpus luteum, which has a constant intrinsic life span(in human about 14 days), seems to be functioning individually.Although the control mechanism of the function and life span of the human corpus luteum has not been fully understood, human menstrual cycle may be controlled by the life span of the corpus luteum.During the luteal phase, anterior pituitary function is not active, and thus the serum LH level is low.Since Pharriss &Wyngarden reported that the ovarian steroid production was affected by prostaglandin (PG), which is a local hormone, it has been elucidated that PGF2α possesses luteolytic activity in such laboratory animals as rat, hamster, pig, cow, sheep, and etc.In human, however, luteolytic effect of PG has not been evident. In this study, a newly developed extra-corpal organ survive device was used for the organ perfusion of "human utero-tubo-ovarian unit"to know luteotropic and luteolytic effects of various chemical substances.Concentrations of progesterone(P)and 20α-hydroxy rogesterone(20α-OH-P)in the perfusate were measured by radio-immuno assays in order to determine the effects of PGS(PG F2α, PG E2, and 16, 16 dimethyl trans Δ2 PGE1 methyl ester(Δ2 PGE1))on the human corpus luteum function.The effects of PGS were compared with those of hCG during the perfusion.The results obtained were as follows:1) After administration of hCG, P concentration was rapidly increased, whereas 20α-OH-P concentration was not.2) After administration of small amount of PGS(PGF2α200μg, PGE2 200μg, Δ2PGE1 20μg), there were no changes in P and 20α-OH-P concentration. Whereas hCG administration followed by PG infusion caused a rapid increase in the P concentration.3) After administration of large amount of PGS(PGF2α500μg, Δ2PGE1 100μg), there were slight decrease in the P concentration, and administration of hCG after PG infusion didn't cause any increase in the P concentration.From these results, it could be concluded that hCG has an evident luteotropic activity while PGF2αand Δ2PGE1 seem to possess luteolytic activity and to inhibit the luteotropic action of hCG in man.
