Experimental and Clinical Studies of Bucolome on the New Born Jaundice

To date it is realized that neonatal icterus closely relates to the function and maturity ofnew born liver.That is to say, neonatal icterus depends on the activity of enzymes concerned bilirubin metabolic pathway such as UDPG-dehydrogenasaen d UDP-gluculonyltransferase.On this view point, the activity of UDPG-dehydrogenasea nd UDP-gluculonyltransferase in the rat liver was measured during development of the fetus and in the neonatal period.Although both enzymes have very low activity in the fetus, activity of the enzymes gradually elevates in the neonatal period.The results obtained indicate these enzymes might be adaptive enzymes hence the possibility of inducing these enzymes by the respective substrate was considered. So, Bucolome (5-n-Butyl-1-cyclic-2, 4, 6-trioxoperhydropyrimidinew) as injected to new born rats, and significant elevation of the UDP-gluculonyltransferasea ctivity was observed after 5 days from the initial injection.Daily administration of Bucolome to pregnant rats for 7 days resulted increase of the enzymes of new born rat liver, while UDPG-dehydrogenaswe as not influenced. On the basis of these experimental results, Bucolome was administrated retrospectively over 10 days daily to 30 pregnant women who have ABO incompatibility.The new born infants treated with Bucolome showed significantly low bilirubin levels compared with non treated infants.
