Design and Function of Proton-Ionizable Cyclam Fluoroionophore for Zinc (II) Ion Sensing in Water

A novel cyclam-based fluoroionophore 1 exhibiting selective fluorescent response for Zn<SUP>2+</SUP> has been designed. The proton dissociation of the pyrenecarboxamide moiety in 1 was promoted by Zn<SUP>2+</SUP> binding, which resulted in the efficient monomer emission response due to direct coordination of nitrogen donor in the carboxamide anion to Zn<SUP>2+</SUP> inside the cyclam binding site. This response mechanism was different from the results observed for 2/Na<SUP>+</SUP> complex possessing the same pyrenecarboxamide function. The apparent binding constant of 1 for Zn<SUP>2+</SUP> was determined to be (2.1±0.2) x 10<SUP>4</SUP> M-1 in 1% 1, 4-dioxane-99% water (v/v) at pH 6.7.
