Study on the mode of inheritance of juvenile onset type insulin dependent diabetes mellitus.

The mode of inheritance of IDDM was analyzed in 165 families. All probands in this reportwere IDDM patients according to the criteria proposed by the WHO Expert Committee. To securea homogenous group of IDDM patients, only patients with onset of the disease before the age of 15years were used.<BR>The families were divided into two subgroups according to the presence or absence of NIDDMpatients in the close relatives of the probands, excluding siblings, as follows: 1) 40 families having NIDDM patients in the close relatives of the probands (Group 1), and 2) 125 families having no NIDDM patients in the family members (Group 2).<BR>The results obtained may be summarized as follows.1) The estimated values of the segregation ratio (Ps) were 0.0400 ± 0.0039 and 0.0072 ± 0.0031for Group 1 and Group 2, respectively. These values were significantly different from either theexpected value of Ps for simple dominant inheritance (0.5) or that for simple recessive inheritance (0.25). The data indicated that the mode of inheritance ofIDDM does not fit the hypothesis ofsimple hereditary modes.<BR>2) The values for the consanguineous marriage rate of the parents of the probands were 0.0556and 0.0183 for Group 1 and Group 2, rspectively. These values do not differ from the consanguineousmarriage rate reported in the Tokyo area (0.0122-0.0711). No effect of consanguineous marriagecould therefore be demonstrated.<BR>3) The mode of inheritance of IDDM observed in the present study does not fit the hypothesisof multifactorial inheritance.
