In vitro studies were carried out to get many microsclerotia (MS) of <I>Calonectria crotalariae</I>, which is causal fungus of soybean black root rot, and to isolate from other propagules. Thirty days after inoculation many MS were formed. in the triple cellophane layers on the modified PDA (1 %agar, 5 %glucose). When upper layer was stripped, mycelial mats were took off together. Only intact MS remained through the middle layer. It was easy to calculate number of MS and to observe germination in the soybean rizosphere. This method should be of use for studying epidemiology of <I>C. crotalariae</I> in soil.
