On the Temperature Condition Suitable for the Detection <I>Erwinia aroideae</I> from Soils by the Carrot Slice Method

When the soft rot bacterium, <I>E. aroideae</I> was detected from soils by the carrot slice method it was ascertained that <I>B. polymyxa</I> was chiefly detected from the rotted carrot slices inoculated with soil suspensions to be tested. It was necessary to establish the conditions to exclude <I>B. polymyxa</I> so as to detect predominantly <I>E. aroideae</I> in the employment of the method. In this study, incubation conditons were examined with special reference to temperature suitable for the detection of <I>E. aroideae</I> from soils by the method. The results obtained are as follows:<BR>1. The strains belonging to E. aroideae could grow in the nutrient broth or sterilized soils at temperatures from 15 & C to 30 & C and showed a vigorous growth at 25 & C-30 & C. Jn <I>B. polymyxa</I>, however, growth did not occur at 15 & C and a vigorus growth was obtained at 30 & C-35 & C. The growth of <I>B. polymyxa</I> in sterilized soils exhibted the same tendencies as in the nutrient broth<BR>2. In the inoculation tests to carrot slices, potato tuber slices and detached petioles of chinese cabbage, the effects of temperatures on the progress of the soft rot lesion caused by <I>E. aroideae</I> differed from those caused by <I>B. polymyxa</I>, that is, <I>B. polymyxa</I> was able to cause lesions rapidly at 30 & C-35 & C and quite slowly below 25 & C. On the other hand, <I>E. aroideae</I> was able to make lesions rapidly even at 20 & C<BR>3. The carrot slices used were apt to be contaminated with <I>B. subtilis</I> when they were incubated at 30 & C-35 & C. The carrot roots bought at the market were contaminated with the soft rot bacteria from August to September.<BR>From the above results it was desirable that in order to detect <I>E. aroideae</I> from soils by the carrot slice method, the carrot slices inoculated with soil suspensions were incubated at 20 & C in the moist chamber for 24hrs-48hrs.
