Studies on the periimplantation development of mouse embryos co-cultured with macrophages.

The periimplantation development and ultrastructual features of mouse embryos co-cultured with macrophages were investigated and compared with those of embryos co-developed in vivo. Pronuclear-stage embryos were collected from superovulated ICR mice and were cultured under four different sets of conditions as follow, 0.3%BSA+BWW, 0.3%BSA+ a -MEM, 0.3%BSA+BWW with guinea-pig peritoneal macrophages and 0.3%BSA+ a -MEM with guinea-pig peritoneal macrophages. Although co-culture with macrophages significantly increased the periimplantation development of mouse embryos, in vitro-developed blastocysts did not implant into the uterus of surrogate female mice unlikely in vivodeveloped blastocysts. Ultrastructual studies revealed a higher rate of mitochondrial degeneration (disrupted mitochondrial cristae, myelinization and reduced electron density) in co-cultured embryos in those in vivo. In conclusion, co-culture of early mouse embryos with macrophages was not available for improving to the implantation rate, presumably resulting in part from accounting for mitochondrial degeneration.
