Sperm immobilization test using cryopreserved sperm preparation.

In our investigations sperm immobilizing antibodies (SIA) were proven to have inhibitory effects not only on the sperm motility with complement, but also on the sperm penetrability through human zonae pellucidae without complement. Therefore, the sperm immobilization test (SIT) should be performed for every infertile couple before starting treatment. It is always a serious problem for clinics to get high quality semen from a particular fertile donor when needed. Therefore, we examined whether or not the cryopreserved sperm preparation could be used for SIT. After swim-up and condensation by centrifugation the semen was cryopreserved in a programmable freezer for three months. The frozen-thawed sperm motility rate gradually reduced from 90% to 30% over the course of cryopreservation. SIT could be performed successfully using a sperm preparation with a motility rate over 30%. Twenty-seven sera were tested five different times over the duration of the cryopreservation. As a result, twenty-four sera were judged as SIA-negative and three as positive by SIT using both fresh sperm and the frozen-thawed sperm preparation which had been cryopreserved up to three months. These results suggest it is pertinent for clinics to use cryopreserved sperm preparation to detect SIA by SIT.
