A study of cellular kinetics in interscapular brown adipose tissue of mouse.

The brown adipose tissue (BAT) is a heatproducing organ in diet-induced thermogenesis and nonshivering thermogenesis. Autopsy from human bodies has revealed the presence of BAT in aged persons as well as in newborns.<BR>This paper describes experiments directed toward cellular kinetics in interscapular BAT (IBAT) of mouse.<BR>1. The change of weight of IBAT in normal mouse with aging<BR>2. The change of weight of IBAT in 20% body weight loss-mouse by starvation with aging<BR>3. The morphodifferential classification of brown fat cells by means of the light and electron microscopic studies<BR>4. The change of index of metabolic activity in IBAT of normal and starving mouse with aging. The index was evaluated by score based on the morphodifferential classification of brown fat cells.<BR>The results are summarised as follows:<BR>1. The ratio of IBAT weight to the body weight decreased with aging in the normal mice.<BR>2. The ratio of IBAT weight to the body weight decreased with aging in the 20% body weight loss-mice.<BR>3. The electron microscopic features showed that the quantities of fat vacuoles were inversely proportional to that of mitochondria in the cytoplasms of brown fat cells. From these findings, the brown fat cells could be morphodif ferentially classified into 7 types.<BR>4. In the normal mice, the metabolic activity index of IBAT decreased significantly with aging. In the 20% body weight loss-old mice above 13 weeks of age, the metabolic activity index of IBAT showed high values in contrast to that of the normal old mice. Concerning the metabolic activity index of IBAT in the young mice under 8 weeks of age, however, there was no appreciable difference between the normal mice and the 20% body weight loss-mice. From the above results, it was concluded that the decrease of the ratio of BAT weight to the body weight and of the metabolic activity index in BAT with aging might result in depression of the diet-induced thermogenesis and the nonshivering thermogenesis in BAT, and that the high metabolic activity index in the 20% body weight loss-old mice above 13 weeks of age implied fat consumption associated with starvation, not direct relation with the natural activity of IBAT.
