Poly [<I>N</I><SUP>5</SUP>- (3-hydroxypropyl) -L-glutamine] membranes crosslinked with 1, 12-diaminododecane were prepared. Changes of the membrane length in aqueous solutions of three kinds of denaturing agents were measured to determine the magnitude and rate of change of the secondary structure in the main chain of the polymer. Sodium dodecyl sulfate (SDS) lengthened the membrane by 32% even in the 10<SUP>-2</SUP> M concentration. The expansion was proved to result from a decrease of α-helix content in the main chain of the polymer according to the infrared absorption spectra. Guanidinium chloride and calcium chloride shortened the membrane by about 10% above the 0.1M concentration. The contraction was proved to result from a increase of α-helix content. Guanidinium chloride and calcium chloride required about 20 seconds to contract the membrane, while SDS required about 60 minutes to expand the membrane. Reversible expansion and contraction behavior of the membrane was observed for all the denaturing agents studied. The permeation rate of vitamin B<SUB>12</SUB> through the membrane was slowed down by adding guanidinium chloride or calcium chloride. The decrease of the permeation rate was mainly due to the decrease of the diffusibility.
