The relation between the mechanical denaturation and the conformation of poly-L-glutamic acid (PGA) in aqueous solution was studied with optical rotatory dispersion (ORD) method. The PGA having α-helical conformation with helix content of 80-90% coagulates by shearing force in the aqueous solutions. These results agree with those of the previous reports.
From the measurements of viscosity and ORD of PGA in these solutions, it is found that the helix content was found to decrease with increasing temperature from 20 to 60°C.
From the IR spectra, the PGA films which were obtained by evaporating these solutions of pH4.0 at room temperature are shown to be of α-helical conformation, but the films obtained at 80°C at the same pH are shown partly of the β-structure.
