Studies in the midgut sucrase types of 39 silkworm varieties which included local varieties, practical original strains and mutants using polyacrylamide gel electrophoresis with sodium dodecyl sulfate followed by activity staining with TTC (SDS-PAGE-activity staining) revealed that some of the silkworm varieties lacked Suc-1 completely. Although Suc-1 was separated into two components (Suc-1a and b) in some strains, neither components were observed to be lacking independently. Linkage analyses performed by using SDS-PAGE-activity staining with marker genes of the respective linkage groups showed that the gene Suc-1 was linked to the 17th group using bts as a marker. Although the activity of the three sucrases was maintained even after the pupation in the midgut lumen, the activity of Suc-1 decreased abruptly 2 days before emergence and disappeared completely in the lumen on a day before emergence. The activities of Suc-2 and 3, in contrast, persisted until the adult stage, though they decreased significantly shortly before emergence.
