The microstructures of silk dyed with acid dye were compared with that of original silk (scoured only) by means of X-ray analysis. Four kinds of specimens prepared were scoured only (Specimen I), scoured in 10-3M HCl solution at 85°C (II), dyed with acid dye at pH 3 and 85°C (III) and dyed with acid dye in the presence of sodium n-dodecylsulfate (SDS) at pH 3 and 85°C (IV). The spacings in all specimens of silk were found to be not essentially altered by treating them in HCl solution and dyeing with acid dye in the absence or in the presence of SDS. The evidence suggested that the taken-up of dye occurred in only amorphous region of silk. By treating the silk in HCl solution the crystallinity increased and degree of orientation decreased. The evidence suggested that the growth of crystallite occurred and the gap between them became rather larger. These structural changes were considered to be favorable for the dye molecule to diffuse into silk and taken up by it. Both the crystallinity and orientation of silk dyed with acid dye in the absence and in the presence of SDS (III and IV) were larger than that of II. The evidence suggested that the dye and SDS taken-up by fibroin molecule in the amorphous region stimulated the molecular orientation of silk.
