In order to gain information on cross-incompatibility in intergeneric hybridization between mulberry and B. kazinoki, germination of pollen on the stigma and development of embryo and endosperm in mulberry crosses and crosses between mulberry and B. kazinoki were examined microscopically. The shape of pollen grains of mulberry and B. kazinoki which were dipped in iron aceto-carmine solution was almost globular, and the pollen grains of mulberry were considerably larger than those of B. kazinoki with a diameter about 1.5 times greater than that of the latter. More than half of the mulberry pollen grains which stuck tightly to the long hairs of the split stigma of mulberry germinated and some of the pollen tubes penetrated into the stigma two hours after pollination. On the other hand, pollen grains of B. kazinoki were easily detached from the hairs of mulberry stigma and did not germinate at all until five days after pollination. In the mulberry crosses double fertilization was observed 10 hours after pollination and the first division of the zygote occurred 48 hours after pollination. Six to nine days after pollination, a considerably large embryo ball was formed through repeated division of the zygote and was surrounded with endosperm which was first formed in the peripheral layer of the embryosac and which grew toward the central part of the embryo-sac. The embryo grew up to the heart shape stage at 10 to 12 days after pollination and to the stage in which cotyledon and hypocotyl could be easily identified 15 days after pollination. In contrast, in the crosses between mulberry and B. kazinoki the division of egg cells and the formation of endosperm were not observed until 16 days after pollination, but the development of the ovule did not differ appreciably from that of the mulberry crosses until four days after pollination. Thereafter great differences could be detected.
