Microsomal S9 fractions from 5th instar larvae of seven stocks of the silkworm (Bombyx mori L.) were compared for their ability to metabolize parent chemicals to mutagens. Drug metabolic activity of the S9 fractions was measured by the His+ Salmonella TA98/silkworm microsome system. In this system, the metabolic activity was assayed as the mutagenicity of the chemical against Salmonella or the increase of His+ independent revertant colonies on minimum glucose media as compared with controls without the microsomal S9 fractions. 2-Acetylaminofluorene (AAF), one of typical indirect mutagens, was used as the test chemical. In Salmonella TA98, AAF is not mutagenic in the absence of the activation systems, but mutagenic in the presence of the systems.The results indicate that the AAF metabolic activity among the stocks was the highest in C108, C108 (kyu), F1 hybrid of Aojuku×C108, Aojuku, Nihonnishiki, rb and Kojiki showed the less activity decreasing in this order. S9 fractions taken from female larvae seemed to have somewhat higher activity for AAF metabolic activations than those of males except for the cases of Nihonnishiki and rb. S9 fractions taken from larvae of C108 and C108 (kyu) showed three times as high activity as liver S9 fractions from rats which had been treated with PCB as an enzyme inducer. Those from F1 hybrid of Aojuku×C108, Aojuku, Nihonnishiki and rb showed almost the same activity as that of rat livers. Only the S9 fraction from Kojiki showed lower activity than that from rat livers. In general, activation system is considerably active in the silkworm larval microsomal S9 fractions.
