Genetic effect of a fungicidal agent, ortho-phenylphenol sodium salt (OPP·Na) on germ-cells of the silkworm (Bombyx mori L.) was investigated. An F1 hybrid from a cross between wild type silkworm strains G108 and Aojuku was used as a material. OPP·Na was supplied from the National Iustitute of Hygienic Science. Toxicity or killing test for a single dose of OPP. Na was measured as the reduction in survival (or adult emergence) ratio. Genetic toxicity was tested by dominant lethal and egg-color specific locus mutation methods.LD50 value on the gram body weight basis was in the range of ca. 100 to 600μg. The results obtained from the dominant lethal test in mid-stage male pupae with OPP·Na indicated no significant increase of dominant lethals above the spontaneous level at 2.2 to 415.3μg/g body weight. It was also observed no evidence for the induction of dominant lethal mutations in pupal oocytes at the concentrations tested ranging from. 1.8 to 182.5μg/g pupal weight. This fungicide at concentrations between 2.2 and 183.5μg/g pupal weight was shown to be non-mutagenic in the specific-locus mutation tests in either pupal sperm or oocytes. It should be noted, however, that OPP·Na can cause high incidence of inf ecundity for mid-stage pupal females but not for pupal males: the number of eggs deposited per female was reduced after treatment with this chemical at a concentration equivalent to ca. one-half of the LD50 dose.
