Midgut trehalase of the silkworm, Bombyx mori has been shown to change its activity and subcellular distribution as well as its kinetic and physical properties during larval-pupal development. To determine whether the changes are specific for metamorphosis, trehalase activities in the midgut were followed from hatching until the last larval ecdysis and also during the larval-pupal development controlled either by an application of juvenile hormone analogue ZR-515 or ecdysterone.Trehalase activity per individual midgut increased exponentially according to larval growth and reached a value some 3000 times larger at the last larval ecdysis than that at hatching, whereas the specific activity remained at almost constant ranges but with a cyclic fluctuation within each instar. The activity was mainly recovered in particulate fractions obtained by centrifugation. Developmental programs altered by ZR-515 or ecdysterone were reflected in the different timing of marked changes in trehalase activity and subcellular distribution; the prolonged larval period resulted in the delayed activity decrease and solubilization of particulate trehalase and vice versa.Consequently the changes of midgut trehalase during larval-pupal development of silkworms are considered to be the changes to reflect the physiological events integrated by the midgut metamorphosis.
