The purpose of the present study was to determine whether ethyl methanesulfonate (EMS) which is a monofunctional alkylating agent and a strong DNA single-strand breaker, has recombinogenic effect on larval oocytes of the silkworm.Three different concentrations (0.25, 0.50 and 1.00%) of EMS solution dissolved in 0.85% NaCl solution were injected into 5th instar female larvae (0.025ml per larva) which were heterozygous in the transhetero type of two egg-color genes, pe (V-0.0) and re (V-31.7). Detection of recombinations was scored by the egg-color method.The frequency of recombinants recovered was very low and not significantly different from that of the control, irrespective of the EMS doses and stages of larvae. It should be noted, however, 0.1% EMS induced mosaic or fractional-body mutations in mature larvae with slightly high frequency compared with the control by the egg-color specific locus method. The incidence of complete or whole-body mutations probably corresponding to double strand DNA changes was not increased. Such being the case, the dose of EMS used in the experiment seemed to be not insufficient for the induction of recombinants. Current evidences indicate that EMS induces a predominant number of single strand breaks (or nick).From these results in the present and previous experiments, recombinogenicity of the alkylating agents may require at least two functional sites. In other words, both strand breakages in the complementary DNA strands may be essentially necessary for the induction of recombinations in prophase I oocytes of the silkworm.
